Mission

The Cellular Immunology and Immune Metabolism Core provides centralized immunology expertise and technologies to support study design, data generation, and interpretation across basic, clinical, and translational research. Through consultation, collaboration, and training, the Core ensures rigorous, reproducible experimentation and meaningful biological insight. Our mission is to enable high-quality research that advances discovery and supports investigators in publication and competitive funding.

 

Core Description

The Cellular Immunology and Immune Metabolism Core (CIMC) supports investigators from the New Orleans metropolitan area including LSU Health Sciences Center School of Medicine, LSU Health Sciences Center School of Dentistry, Tulane University School of Medicine, Ochsner Health System, and Xavier University.  The Core provides expertise in sample processing and instrument operation, as well as consultation on experimental design, troubleshooting, data analysis, and interpretation of results.

In addition, the Core offers group and individual training sessions, workshops, and seminars for investigators and trainees.

Core instrumentation is available 24/7 to trained users, apart from the cell sorter. All equipment is secure with user-specific access, allowing for usage tracking, performance monitoring and proper operation.

BD FACSymphony S6 (2024) 25-paramiter cell sorter equipped with five lasers: 349nm; 405nm; 488nm, 561nm and 633nm. Sorter is used for isolation of specific cells population based on their cellular markers. It can sort up to six different cell populations into 5 ml tubes. Additionally, cells can be sorted to 6-, 24-, 48-, 96-, 384 well plates or slides. Operated only by Core personnel.

Beckman Coulter 10 color Gallios flow cytometer, purchased in 2010. Gallios is equipped with 3 lasers: 405nm; 488nm and 638nm. Available to be operated by a user after training.

BD FACSymphony A3; 25-parameter flow cytometer, equipped with five lasers: 355nm; 405nm; 488nm, 561nm and 633nm. Available to be operated by a user after training.

Amnis ImageStreamX Mark II combines flow cytometry with fluorescent microscopy technology. This flow cytometer is equipped with a camera, which takes a picture of every cell passing through the flow cell using 20x, 40x or 60x magnification lenses. Equipped with four lasers: 405nm; 488nm, 561nm and 633nm. This configuration, besides regular cell phenotyping, allows analysis of cell-to-cell interactions, cell signaling, internalization and co-localization, translocations, shape change and chemotaxis and nanoparticles analysis. Available to be operated by a user after training.

The Incucyte SX5 Live-Cell Analysis System is a real-time cell monitoring and surveillance system that automatically captures high resolution fluorescent and bright field images. It analyzes images of living cells around-the-clock for days, weeks, or months, while cells remain undisturbed inside a standard tissue culture incubator. The stage accommodates up to six multi-well plates, which are scanned automatically according to the investigator-defined schedule. Incucyte enables real-time monitoring of a variety of live cell assays including viability, differentiation, proliferation, migration, invasion, metabolism, neurite outgrowth and a wide range of phenotypic cell-based assays. Available to operate by user after training.

Cytation 5 BioTek combines automated microscopy and conventional microplate reader. Magnifications 4x, 10x, 20x, 40x and 60x in fluorescence, high contrast brightfield, color brightfield, and phase contrast. Fully automated microscopy tasks, from slide scanning to time-lapse live cell assays. The multimode detection modules allow to read fluorescence, luminescence, and UV-Vis absorbance. Gen5 software provides complete control over all imaging and data analysis.

Seahorse XFe96 bio analyzer (previously named Seahorse) measures real time cell respiration in 96 well plate. It is used to perform assays like “Glycolysis Stress”, MitoStress, “ATP Rate” and many others. Our XFe96 unit is placed inside the hypoxia chamber, which allows creating different oxygen conditions. XFe96 is equipped with Cytation 1 microscope for cell counting to normalize the results. Available to be operated by user after training. Kits and reagents can be found here.

Luminex -MagPIX. It can measure up to 60 analytes /sample in a single well. MagPIX technology is a beads-based versatile multiplexing platform performing quantitative and qualitative analysis of proteins and nucleic acids in a variety of sample matrices. The IVIS® SpectrumCT by Perkin Elmer is a preclinical in vivo imaging system, which allows simultaneous molecular and anatomical non-invasive imaging of small live animals. It is used in monitoring disease progression, cell trafficking, and gene expression patterns. IVIS Spectrum CT offers 2D and 3D imaging capabilities and includes integrated low-dose microCT. Available to operate by user after training.

Malvern Panalytical Inc.-NanoSight NS300. It is a system for detection and measuring the size and concertation of extracellular vesicles, EV. Equipped with 532 nm laser. Milteny gentleMax Octo dissociator with heaters for preparation of single cell suspension from tissues and tumors. (Not in iLab) available to everyone without training. 

Eco Revolve small fluorescent microscope. Operated by a user after training.

All equipment is under a service contact, and all calibration is performed daily, which allows instruments to be well maintained and in case of malfunction fast repair.

The following analysis software are available: Kaluza, Flowjo, ModFit, Ideas, Wave, Excel, Graph Pad software.

Please contact the Core before planning the experiment, to make sure fluorophores are compatible with the systems in the Core.
